Ships from United States.Cons: I purchased this converter to digitize 10 years-old (but well maintained) 35 mm negatives. After converting a few images and trying different settings, I was very unhappy with the quality of images. Almost none of the colors matched the original colors. I understand age-degradation but I still expected a reasonable representation which I was unable to achieve after trying different resolutions and settings. I was also forced to upload the images after about 10 captures which was a hassle.http://www.fiacasyfutones.com.ar/userfiles/endress-hauser-promag-53-flow-meter-manual.xml The menu options are limited (no color-adjustment just the light-control) and I didn't find anyway to save my settings so I had to reset options after each start-up. Overall Review: The search goes on for a better quality converter. Did you find this review helpful. Cons: Mine came with a small black spot on the viewer that the cleaner could not get rid of. The video cable or the jack had a short in it that caused some minor probs. If using slides like Kodachrome Transparancy circa 1950 the slides are larger than the unit can handle (hence you loose some pix at the edge of slide). The frames for holding the slides are not easy to open. The stops for the slide holder do not always center on the slide (the first slide in holder the worst). The printed manual is not of a size that is easy to see (especially the icons). Overall Review: Even though this product is challenging because of it's liittle problems it is an effective product. Chris Higgins 3.0 out of 5 stars The scans are cropped (missing the edges), and they are low resolution. If you seek a serious film scan, look for a much higher-end scanner. The main point here is that this slide scanner is decent but not great. If you want archival quality, don't waste your time. Get something far better.I had tried scanning them earlier with an HP scanner, but the quality was poor unless I increased the pixel count, and then it took 3-5 minutes to scan a slide. With the VuPoint, the process was so much easier and the actual scanning of each slide took less than 5 seconds. But, the best part was the quality, which was as good or better than with my HP scanner. As with both experiences, some slides need to be lightened. I've used my Adobe Photoshop software, so I can't speak for the software that comes with the VuPoint. We can now share memories of events, trips, special people with our family. For the price, the VuPoint has exceeded my expectations. You have to place the slides 3-at-a-time in a tray and then run them through the machine, one at a time. This work is slow and tedious, but the resulting images are very sharp if viewed on a computer screen or other device and they are JPEGs, so they can easily be emailed or burned on a DVD, with overall excellent results. My advice is to set this up where you can leave it set up and you can go back to it off and on, over the course of the day and run a few slides through at a time. To sit there for hours and do it can get really tedious. If you can afford to do so, hire some little kid to do it for you. Any average ten-year-old can master this machine a ten minutes. Simply insert it into this device and you're good to go. It comes with a cord that allows you to send the images directly to a tv screen,so you can create an old-fashioned slide show, but I don't recommend doing it. At that size, the images are not very sharp and the process is much too slow and tedious. Your audience will fall asleep (or slip away in the dark.).Not one of those things you can do by rote, assembly line movements. The effort required close attention to details, and the set up of the equipment was not easy. One problem is that the power cord is short, and doesn't allow any movement of the converter away from the electric socket. My primary complaint is that the unit stopped working after about a hundred conversions, and the unit was returnable to Amazon after only 30 days. I did not immediately try to use the converter after I received it, so the 30 days had nearly expired when the unit stopped working. I notified amazon of the problem and requested return instructions, but I never heard back from them. VuPoint was very helpful in replacing my unit.No buzzing, waiting. I have hundreds of slides which I don't want to keep - so this converter works well for my purposes. The slide carrier is poorly designed - and because of that it doesn't fit the slide mount very well and doesn't close completely. Therefore it's hard to put the slide carrier all the way through. The slide carrier is strictly and push and pull operation. I am able to use just one half of the carrier - so instead of loading all 4, I load two. I have a photo editing software which I use to process the scans afterwards - so if you don't have that, you might not like this converter. If I have a slide that is really special, I would not rely on this scanner to make a copy worth keeping. The slow part is finding your negatives which may be difficult, and putting them in the holder, which is easy. It is designed for 35mm negatives or standard slides. It does the job I wanted, which was to inventory my thousands of negatives. I've just begun on that task, and am appreciative of a device I can use with ease. Once I set it for negatives, I only have to open the lid and it is ready to do more of the same. I can add the pictures put on my SD card to my computer and print from there. More importantly, I can search for and find negatives of the significant photos I want to keep. I want to find a place to print high-quality enlargements, from the original negatives, of only the 3 or 4 pictures I'm especially looking for. But, alas not so easily done. It takes 3 or 4 attempts to transfer to the computer. I've been able to retrieve 4 images so far. And one is led to believe that you can transfer 4 slides at a time. What I've found is that you can transfer only one at a time; the other just sit there or disappear; and for the ones that succeed it takes 2 to 3 attempts; totally frustrating to say the least. It has met my needs and was easy to set up and use.Page 1 of 1 Start over Page 1 of 1 In order to navigate out of this carousel please use your heading shortcut key to navigate to the next or previous heading. Following the instructions was hard at first but after using it for a short time it became eaiser. The only problem is that the instructions tell you to clean the lens periodically but it don't tell you where it is or how to get to it. Pros: Uses standard SD card, good color reproduction, scans mounted 35mm slides into standard jpeg file. It is slow and rather awkward to use, especially if you have hundreds of slides to convert. Some chromosomal aberrations, in particularly in the telomeric region, are difficult to identify due to limited resolution of chromosome specific banding pattern. Similarly, conventional cytogenetics is difficult to apply to study chromosomes in single cells like blastomere (as in preimplantation diagnosis), polar body and gamete (oocyte and sperm). Conventional cytogenetics is possible only in dividing cells, has limited resolution (4 Mb, Rimion et al. 1996, however, 1-2 Mb can be reached in long chromosome preparation) and have no functional information. These factors have led investigators to seek newer approaches for identifying chromosomal QFPCR although can not localize anatomically, it can trace parental origin of chromosomes as well as can provide information on maternal contamination in prenatal diagnostic situation. High sensitivity, specificity, speed and the ability to provide information on single cell have made these newer molecular approaches a powerful tool in modern cytogenetics. An understanding of these genetic changes provides knowledge for oncology researchers that will lead to significantly improved detection methods, therapies and disease prevention. It highlights the impact that these approaches have on disease management along with the experience of authors. FISH In-situ Hybridization (ISH) allows specific nucleic acid sequences to be detected in morphologically preserved chromosomes, cells or tissue sections.T echnique was developed initially by Pardue and Gall (1969) and John et al. (1969) independently. Use of ISH was limited initially due to nonavailability of probes particularly nonradioactive. A number of developments in 1980s viz.Furthermore, it opens new opportunities for combining different labels in one experiment. In the direct method, the detectable molecules e.g., fluorescein isothiocyanate (FITC), T exas Red isothiocyanate (TRITC), rhodamine, etc are bound directly to the desired nucleic acid sequence (probes) so that probe- target hybrids can be visualized under microscope immediately after hybridization reaction. Indirect procedures require the probe to contain a reporter molecule (e.g., biotin: Langer et al. 1981 and digoxigenin: Heiles et al. 1988) that can be detected by affinity cytochemistry with fluorescent dyes. This present state of ISH is known as Fluorescent ISH i.e., FISH. It provides a direct link between microscope and DNA sequence. FISH probes can be either cloned DNA sequences or generated by PCR from flow-sorted chromosomes or micro-dissected chromosomes or even from whole genome. Cloned DNA sequences may be as alphoid sequence (171 bp repeats), telomeric sequence (5-6 bp repeats), subtelomeric sequence, unique locus specific sequence, etc. Cloned sequences are obtained as PCR product, plasmid ( insert), cosmid ( AC ( BAC ( AC ( Labeled (by nick translation or PCR) nucleic acid sequence (i.e., probe) is annealed to the sample after denaturation (both probe and test DNA). After washing to remove un- hybridized probes, the location of the hybrids is visualized by fluorescence microscopy (a detection system is necessary for indirectly labeled probes). Sensitivity-specificity of probes varies with type of probe as well as indications. However, it may be much lower for monosomy diagnosis as false monosomy may be as high as 10 (Poddighe et al. 1991). Sensitivity is further decreased in formalin fixed tissue where it may be as low as 50. Conventional FISH T ypes of Probes DNA Probes Chromosome Paint Whole Chromosome Paints (WCP; figure 1): These probes, as the name suggests, paint the entire length of a metaphase chromosome. The chromosomal fragments can be produced in the somatic cell hybrids as the consequence of in vitro rearrangements. Hamster-human somatic cell hybrids are prone to have rearrangements. These probes also can be generated by micro-dissection and PCR amplification protocols as in WCP. 1998). Polymerase Chain Reaction (PCR) constitutes a simple way to generate and label centromeric (alphoid) probes at low cost (W eier et al. 1991). T wo types of human DNA source can be used: a. Somatic hybrid cell lines containing a unique human chromosome. The specific amplification of the human subset of alphoid DNA is realized with a primer pair specific for the consensus region of human alpha satellite sequence. b. T otal Human DNA. This time, a primer pair Figure 2 Showing conventional multicolor (three) FISH using chromosome 1 (centromeric; yellow), chromosome X (centromeric; green; FITC) and chromosome Y (q heterochromatic; red; rhodamine) probes. Although the basic or ganization is always characterized by monomers of about 170bp tandem repeat (few hundred to few thousand times), the nucleotide sequence and monomeric organization may be considerably divergent from one chromosome to another. Therefore, under appropriate stringency conditions, an alphoid clone may recognize a single chromosome. Chromosome-specific subsets have been described for most of the human chromosomes. This also can be produced by PCR amplifications with specific primers (Dunham et al. 1992, Lengauer et al. 1994, Hasegawa et al. 1995, Dupont et al. These probes can be labeled during the PCR reaction (or by separate nick translation) by direct incorporation of modified dUTP. Alphoid probes are very useful for beginners, since the target sequence is usually very large (1-2Mb), so that the FISH signal is strong (figure 2). Alphoid probes are very useful in clinical cytogenetics in particular prenatal and preimplantation diagnosis, polymorphism detection, construction of centromere-based genetic linkage maps, identification of aneuploidies in interphase nuclei of tumor cells, understanding chromosome phenomena such as non-homologous recombination (figure 3), Robertsonian translocations, etc.
